    Hedgehog signaling patterns the outgrowth of unpaired skeletal appendages in zebrafish

    Background: Little is known about the control of the development of vertebrate unpaired appendages such as the caudal fin, one of the key morphological specializations of fishes. Recent analysis of lamprey and dogshark median fins suggests the co-option of some molecular mechanisms between paired and median in Chondrichthyes. However, the extent to which the molecular mechanisms patterning paired and median fins are shared remains unknown.Results: Here we provide molecular description of the initial ontogeny of the median fins in zebrafish and present several independent lines of evidence that Sonic hedgehog signaling emanating from the embryonic midline is essential for establishment and outgrowth of the caudal fin primordium. However, gene expression analysis shows that the primordium of the adult caudal fin does not harbor a Sonic hedgehog-expressing domain equivalent to the Shh secreting zone of polarizing activity ( ZPA) of paired appendages.Conclusion: Our results suggest that Hedgehog proteins can regulate skeletal appendage outgrowth independent of a ZPA and demonstrates an unexpected mechanism for mediating Shh signals in a median fin primordium. The median fins evolved before paired fins in early craniates, thus the patterning of the median fins may be an ancestral mechanism that controls the outgrowth of skeletogenic appendages in vertebrates

    An X-ray and Optical Study of Matter Distribution in the Galaxy Cluster A 2319

    A new analysis of velocity distribution, optical photometry and X-ray surface brightness from ROSAT PSPC data of the galaxy cluster A 2319 is presented. The temperature profile derived from ASCA data (Markevitch et al.,1996) is taken into account. A method to check the hydrostatic model in the presence of a temperature gradient is proposed. Consistency of the hydrostatic isothermal model and the explanation of the "beta-discrepancy" are discussed. Galaxy and gas density profiles of the main component A 2319A are derived, allowing for the effect of the secondary component A 2319B. The inadequacy of a polytropic model, which would produce a binding mass decrease with respect to the isothermal beta-model, is discussed. A simple interpolation of the temperature profile provides instead an increase of the binding mass and a lower baryon fraction thus mitigating the "baryon catastrophe". Assuming as typical the value f_b ~ 0.2, a comparison with the most recent estimate of Omega_b(nucl) implies for the cosmological parameter Omega_o less than 0.4.Comment: 7 pages, 2 tables, 8 figure

    Modeling of Wnt-mediated tissue patterning in vertebrate embryogenesis

    During embryogenesis, morphogens form a concentration gradient in responsive tissue, which is then translated into a spatial cellular pattern. The mechanisms by which morphogens spread through a tissue to establish such a morphogenetic field remain elusive. Here, we investigate by mutually complementary simulations and in vivo experiments how Wnt morphogen transport by cytonemes differs from typically assumed diffusion-based transport for patterning of highly dynamic tissue such as the neural plate in zebrafish. Stochasticity strongly influences fate acquisition at the single cell level and results in fluctuating boundaries between pattern regions. Stable patterning can be achieved by sorting through concentration dependent cell migration and apoptosis, independent of the morphogen transport mechanism. We show that Wnt transport by cytonemes achieves distinct Wnt thresholds for the brain primordia earlier compared with diffusion-based transport. We conclude that a cytoneme-mediated morphogen transport together with directed cell sorting is a potentially favored mechanism to establish morphogen gradients in rapidly expanding developmental systems

    Dwarf galaxy populations in present-day galaxy clusters: I. Abundances and red fractions

    We compare the galaxy population in the Virgo, Fornax, Coma and Perseus cluster to a state-of-the-art semi-analytic model, focusing on the regime of dwarf galaxies with luminosities from approximately 10^8 L_sun to 10^9 L_sun. We find that the number density profiles of dwarfs in observed clusters are reproduced reasonably well, and that the red fractions of model clusters provide a good match to Coma and Perseus. On the other hand, the red fraction among dwarf galaxies in Virgo is clearly lower than in model clusters. We argue that this is mainly caused by the treatment of environmental effects in the model. This explanation is supported by our finding that the colours of central ("field") dwarf galaxies are reproduced well, in contrast to previous claims. Finally, we find that the dwarf-to-giant ratio in model clusters is too high. This may indicate that the current model prescription for tidal disruption of faint galaxies is still not efficient enough.Comment: 20 pages, 10 figures. Accepted by MNRAS.     Regionalizing the infrastructure turn : a research agenda

    An interdisciplinary ‘infrastructure turn’ has emerged over the past 20 years that disputes the concept of urban infrastructure as a staid or neutral set of physical artefacts. Responding to the increased conceptual, geographical and political importance of infrastructure – and endemic issues of access, expertise and governance that the varied provision of infrastructures can cause – this intervention asserts the significance of applying a regional perspective to the infrastructure turn. This paper forwards a critical research agenda for the study of ‘infrastructural regionalisms’ to interrogate: (1) how we study and produce knowledge about infrastructure; (2) how infrastructure is governed across or constrained by jurisdictional boundaries; (3) who drives the construction of regional infrastructural imaginaries; and (4) how individuals and communities differentially experience regional space through infrastructure. Analysing regions through infrastructure provides a novel perspective on the regional question and consequently offers a framework to understand better the implications of the current infrastructure moment for regional spaces worldwide

    A Search for Jet Handedness in Hadronic Z0Z^0 Decays

    We have searched for signatures of polarization in hadronic jets from Z0qqˉZ^0 \to q \bar{q} decays using the ``jet handedness'' method. The polar angle asymmetry induced by the high SLC electron-beam polarization was used to separate quark jets from antiquark jets, expected to be left- and right-polarized, respectively. We find no evidence for jet handedness in our global sample or in a sample of light quark jets and we set upper limits at the 95% C.L. of 0.063 and 0.099 respectively on the magnitude of the analyzing power of the method proposed by Efremov {\it et al.}Comment: Revtex, 8 pages, 2 figure
